This was my guess as well, I always figured the main six were in their mid to late twenties and that in "The Last Problem" they were in their late forties-early fifties

Granny Smith was present for the founding of Ponyville. And Ponyville is over 400 years old. Earth ponies can be very long-lived
Actualy, thinking about it, it gets interesting when you realize that Sunset Shimmer is likely around the same age as Twilight Velvet and that at the beginning of the show Spike is around 18
